Peroxynitrite has been reported to react with L-tyrosine to produce 3-nitro-L-tyrosine (NO2-Tyr), which appears to be a suitable marker for ONOO- mediated tissue damage. The NO2-Tyr standards are analyzed in a dual cell configuration with a detection limit of 0.5 nmol/L. As conversion in the first flow cell increases with lowering the flow rate, a micro LC (1mm) system is required. The ALEXYS system is optimised for analysis of small sample volumes and best detection performance.

System configuration

For the analysis of nitrotyrosine we selected the VT03 electrochemical cell. This cell not only has an excellent reproducibility but also the best possible detection sensitivity. We routinely inject 2-5 µL with a maximum sample use of only 10 µL. Therefore, we selected a micro version of our AS 110 autosampler together with a micro LC 110 pump. We use a 1 mm ID column at 50 µL/min as a standard.
